Overview

Butyrophilin subfamily 1 member A1 (BTN1A1) is a membrane-associated glycoprotein belonging to the immunoglobulin superfamily. It plays a crucial role in milk fat droplet secretion and acts as an immune checkpoint molecule, suppressing T-cell activation. It is highly expressed in several solid tumor types, where it contributes to immune evasion. It has also been implicated in the pathogenesis of multiple sclerosis.

Mechanism of action

BTN1A1 blockade enhances antitumor immunity by increasing immune cell infiltration into tumors. It inhibits proliferation of CD4+ and CD8+ T-cells activated by anti-CD3 antibodies; reduces IL2 and IFNG secretion from these cells.
