Overview

Butyrophilin subfamily 3 member A3 (BTN3A3) is a type I membrane glycoprotein of the immunoglobulin superfamily encoded on human chromosome 6 within the MHC-associated butyrophilin gene cluster. Its ectodomain forms V-shaped homodimers and is structurally homologous to the B7 superfamily; BTN3A-specific antibodies bind distinct epitopes on the IgV domain and can either stimulate or inhibit Vγ9Vδ2 T-cell responses, indicating a role for BTN3A molecules in modulating γδ T-cell activation in infection and cancer contexts. Mechanism of action

Agonist anti-BTN3A antibodies can mimic phosphoantigen-mediated activation of Vγ9Vδ2 T cells by binding BTN3A IgV domain epitopes, modulating T-cell activation signaling. Antagonist anti-BTN3A antibodies can inhibit Vγ9Vδ2 T-cell reactivity by binding distinct BTN3A epitopes and blocking activation.
