Overview

C-C chemokine receptor-like 2 (CCRL2) is a non-signaling, seven-transmembrane domain protein structurally related to chemokine receptors but does not induce classical G protein-coupled receptor signaling[2][3][4]. It is best classified as an atypical chemokine receptor (ACKR5) and is involved in immune regulation, particularly by binding chemerin (RARRES2) and possibly acting as a ligand-presenting molecule to functional chemerin receptor-expressing cells (CMKLR1)[3]. CCRL2 is highly expressed on neutrophils, monocytes, dendritic cells, macrophages, endothelial and epithelial cells, and is upregulated during inflammatory responses[2][3][4]. Its role includes modulating the availability and presentation of chemotactic factors during inflammatory and immune settings but it is not itself a classical signaling receptor; its loss or altered function has been implicated in diseases such as rheumatoid arthritis, allergic responses, and experimental autoimmune encephalomyelitis[3][4]. There are no approved drugs directly targeting CCRL2, but preclinical data support its relevance in immune and inflammatory diseases.
