Overview

C-C chemokine receptor type 10 (CCR10) is a G protein-coupled receptor (GPCR) that serves as a critical regulator of immune cell trafficking to epithelial tissues, including the skin and mucosal surfaces. It primarily interacts with two ligands: CCL27 (CTACK), which is predominantly expressed in the skin, and CCL28 (MEC), which is found in various mucosal tissues. CCR10 is expressed on specific subsets of immune cells, such as cutaneous lymphocyte antigen (CLA)-positive memory T cells and IgA-secreting plasma cells, facilitating their homing to sites of inflammation or homeostatic maintenance. In disease contexts, the CCR10-CCL27/CCL28 axis is implicated in the pathogenesis of inflammatory skin disorders like psoriasis and atopic dermatitis, as well as in the progression and metastasis of cancers such as melanoma and multiple myeloma. While no CCR10-targeted therapies are currently approved, research is focused on developing small molecule antagonists like BI-6901 and monoclonal antibodies to block pathological cell recruitment, as well as utilizing CCR10 expression to enhance the homing of adoptive cell therapies like CAR-T cells to solid tumors.
