Overview

C-C chemokine receptor type 2 (CCR2), also known as CD192, is a G protein-coupled receptor (GPCR) primarily expressed on monocytes/macrophages and lymphocytes. It plays a central role in mediating the migration of these immune cells to sites of inflammation by responding to specific chemokines. CCR2 is encoded by the *CCR2* gene in humans, which produces two main isoforms through alternative splicing: CCR2A and CCR2B. The "type 2b" designation refers specifically to the CCR2B isoform. The primary function is mediating chemotaxis—the directed migration—of immune cells toward higher concentrations of its ligands at sites of tissue injury or infection. Due to its central role in inflammatory cell recruitment and disease progression across multiple organ systems—including liver fibrosis/cancer—CCR2 is considered an attractive therapeutic target for drug development efforts aimed at modulating immune responses or reducing pathological fibrosis/tumor growth.

Mechanism of action

Ligand binding activates G-protein signaling pathways leading to calcium mobilization and inhibition of adenylyl cyclase activity. Promotes T-cell differentiation into Th17 cells via cytokine regulation. Regulates desensitization/internalization via clathrin-mediated endocytosis after activation.
