CCR3 is a G protein-coupled receptor that binds to several CC-type chemokines, including CCL11 (Eotaxin), CCL26 (Eotaxin-3), CCL7 (MCP-3), CCL13 (MCP-4), CCL5 (RANTES), CCL15, CCL24, and CCL28. It is highly expressed on eosinophils and basophils, as well as Th1 and Th2 lymphocytes and airway epithelial cells. CCR3 plays a critical role in mediating the migration of eosinophils and other inflammatory cells to sites of allergic inflammation and parasitic infection. It also functions as an entry co-receptor for HIV-1. Due to its role in allergic diseases and potential involvement in cancer metastasis, CCR3 is a target for therapeutic intervention.
Ligand binding activates intracellular signaling pathways via heterotrimeric G-proteins and other effectors such as β-arrestins and JAK-STAT pathways.
