Overview

C-C motif chemokine ligand 15 (CCL15) is a small secreted cytokine of the CC chemokine family, primarily produced by the liver, small intestine, colon, and certain immune cells (e.g., macrophages, leukocytes)[2][3]. It functions as a chemotactic factor, attracting immune cells such as monocytes, T lymphocytes, and, to a lesser extent, neutrophils, largely by binding to and activating the chemokine receptors CCR1 and CCR3[2][3][4]. The gene encoding CCL15 is located on human chromosome 17 and undergoes proteolytic processing, resulting in multiple active N-terminal truncations with differential signaling and immune effects[1][4]. CCL15 plays a central role in inflammation and the immune response by directing leukocyte trafficking to sites of tissue injury or infection. Dysregulation of CCL15 or its receptors is implicated in chronic inflammation and can contribute to pathologies such as cancer, infection, and other immune-mediated diseases[2][3]. No approved drugs specifically target CCL15, but its role in CCR1-mediated pathways makes it a therapeutic target for anti-inflammatory and anti-cancer strategies[1][3][4].

Mechanism of action

As an agonist: CCL15 binds to and activates chemokine receptors such as CCR1 (and CCR3), inducing G protein-coupled signaling that triggers immune cell migration and inflammatory cascades[1][2][4]. Antagonists (for the receptor): Blockage of receptor prevents CCL15-mediated immune cell recruitment and inflammation.
