C-C motif chemokine ligand 4-like 2 (CCL4L2) is a member of the CC chemokine cytokine family, acting as a non-allelic copy of CCL4 with only a single amino acid difference[2]. CCL4L2 is involved in immune regulation and inflammation, primarily by encoding a chemotactic signaling protein that recruits and activates immune cells such as pro-inflammatory (M1) macrophages at sites of injury or disease[2]. Its expression is increased in inflammatory states and contributes to chronic inflammation and tissue damage, notably in tendinopathy and autoimmune diseases. CCL4L2 impacts cell signaling that inhibits tendon stromal progenitor cell (TSPC) differentiation, promotes macrophage-driven inflammation, and disrupts tissue repair processes. It is considered a potential therapeutic target for inflammatory disease modulation and a candidate biomarker in conditions such as tendinopathy and other chronic inflammatory diseases[2].
