C-C motif chemokine receptor 5 (CCR5) is a G protein-coupled receptor primarily expressed on the surface of T cells, macrophages, dendritic cells, eosinophils, and microglia. It binds the chemokines CCL3, CCL4, and CCL5, mediating immune cell chemotaxis and inflammatory signaling[1][2]. Importantly, CCR5 is a major co-receptor for entry of macrophage-tropic (M-tropic) HIV-1 strains into CD4+ T cells. Individuals homozygous for the CCR5 Δ32 mutation lack functional cell-surface CCR5 and are highly resistant to HIV-1 infection, which has directed the development of CCR5 antagonists as HIV therapeutics[2][3]. CCR5 is also implicated in cancer cell migration and metastasis, inflammatory diseases, and serves as a toxin entry receptor for certain pathogens[1][3].
