Overview

C-C motif chemokine receptor 9 (CCR9) is a class A G protein-coupled receptor primarily expressed on developing thymocytes, intestinal lymphocytes, and other immune cells[1][2][3][4]. It binds specifically to the chemokine ligand CCL25 (also known as TECK), mediating chemotaxis and tissue-specific homing of T lymphocytes, particularly to the small intestine and thymus[1][2]. CCR9 regulates thymocyte migration, maturation, and gut immune surveillance, and plays a central role in many inflammatory and autoimmune diseases including inflammatory bowel disease, rheumatoid arthritis, and a range of cancers[1][2]. Its expression and functional activity contribute directly to inflammation, immune cell trafficking, cell proliferation, and metastasis, making it a validated target for therapeutic intervention in these diseases. Drugs such as vercirnon and other CCR9 antagonists have been investigated to block pathogenic immune cell recruitment and signaling via this receptor[1][4]. Therapeutic challenges include immune suppression and maintaining physiological immune function, especially in the gut.
