Overview

c-Jun N-terminal kinases (JNKs) are a subgroup of the mitogen-activated protein kinase (MAPK) family, also known as stress-activated protein kinases (SAPKs). JNKs play a central role in cellular responses to various stress stimuli. They regulate diverse biological processes such as gene expression, neuronal plasticity, cell proliferation and differentiation, apoptosis, inflammation, and cellular senescence. Dysregulation of the pathway has been linked to numerous diseases, including neurodegeneration, metabolic disorders, cancer, and immune disorders, making it an attractive therapeutic target. JNKs are encoded by three genes: MAPK8 (JNK1), MAPK9 (JNK2), and MAPK10 (JNK3). Selective inhibition or modulation of particular isoforms represents a potential therapeutic strategy.
