Overview

c-Jun N-terminal kinase 1 α2 isoform (JNK1α2) is a variant of the JNK1 protein, part of the mitogen-activated protein kinase (MAPK) family involved in transducing extracellular signals to the cell nucleus. The α2 splice variant of JNK1 is primarily localized to the cytoplasm and lacks autophosphorylation activity, distinguishing it from other JNK isoforms such as JNK2α2. JNK1 plays a key role in controlling processes such as apoptosis, gene expression, and cellular stress responses. Its activity is regulated by interaction with scaffold proteins like JIP1, and it is a therapeutic target in diseases including cancer, inflammatory disorders, neurodegeneration, and cardiovascular conditions. SP600125 is a known ATP-competitive inhibitor that selectively targets JNK1 activity. The JNK pathway, including JNK1α2, is tightly regulated and impacting its function can have broad cellular consequences.
