Overview

JNK1 and JNK2 are members of the c-Jun N-terminal kinase (JNK) family, which belong to the mitogen-activated protein kinase (MAPK) superfamily. These kinases are serine/threonine protein kinases that play crucial roles in transducing signals from various extracellular stimuli to regulate gene expression, cellular proliferation, differentiation, survival, apoptosis, immune responses, and stress responses. They are activated by dual phosphorylation on threonine and tyrosine residues within a TPY motif, primarily by upstream kinases MKK4/7. JNKs regulate apoptosis, cell proliferation/differentiation, and autophagy. They are implicated in various diseases, including cancer, neurodegeneration, and inflammation.
