Overview

The c-MET receptor tyrosine kinase, also known as the hepatocyte growth factor receptor (HGFR), is a transmembrane receptor that binds hepatocyte growth factor (HGF). Upon HGF binding, c-MET dimerizes and autophosphorylates, activating downstream signaling pathways including PI3K/AKT, Ras/MAPK, and JAK/STAT, which promote cell proliferation, survival, and motility. Aberrant c-MET activation due to mutations, amplification, or overexpression is implicated in various cancers, making it a significant therapeutic target. Small molecule inhibitors targeting the ATP-binding site of c-MET have been developed and are used clinically.
