C-Myc-binding protein (MYCBP), frequently referred to as AMY-1, is a protein that acts as a critical coactivator for the MYC proto-oncogene [1]. It specifically binds to the N-terminal transactivation domain of MYC, thereby stimulating MYC-mediated transcription of genes essential for cell growth and division [4]. In addition to its nuclear role, MYCBP is localized to the mitochondria and cytoplasm, where it interacts with A-kinase anchoring proteins (AKAPs), potentially influencing cAMP-dependent signaling pathways [3]. Because MYC is a central driver in many human malignancies, MYCBP has emerged as a significant therapeutic target; its overexpression is linked to increased malignancy and poor clinical outcomes in cancers such as breast and gastric carcinoma [2]. Therapeutic strategies targeting MYCBP include the use of antisense oligonucleotides or siRNA to degrade MYCBP mRNA, as well as small molecules designed to disrupt the MYC-MYCBP protein-protein interaction [1]. However, challenges remain regarding the potential for systemic toxicity, given the fundamental role of MYC signaling in normal regenerative tissues [4].
