The c-Myc peptide tag is a widely used epitope tag derived from the human c-Myc proto-oncogene, specifically representing the amino acid sequence EQKLISEEDL (residues 410-419) (Evan et al., 1985, Mol Cell Biol). It is primarily utilized as a molecular tool to facilitate the detection, purification, and localization of recombinant proteins in various experimental systems (Terpe, 2003, Appl Microbiol Biotechnol). Because the tag is small and typically non-interfering, it can be fused to either the N- or C-terminus of a protein of interest without significantly altering the protein's native function or folding. In laboratory settings, high-affinity monoclonal antibodies, such as the 9E10 clone, are employed to recognize the tag for applications including Western blotting, immunoprecipitation, and flow cytometry (Hilpert et al., 2001, Protein Eng). While the full-length c-Myc protein is a critical transcription factor frequently dysregulated in cancer, the c-Myc peptide tag itself has no known biological function or therapeutic relevance in humans. Consequently, it is classified as a laboratory reagent rather than a drug target or a biomarker for disease (UniProt P01106). The tag's utility is strictly limited to the research and diagnostic development space, with no direct role in clinical pharmacology.
