Overview

ROS1 is a receptor tyrosine kinase involved in epithelial cell differentiation and regionalization. Somatic rearrangements leading to fusion proteins containing the ROS1 kinase domain are oncogenic drivers, particularly in non-small cell lung cancer. These fusions activate downstream signaling pathways like RAS-MAPK, PI3K-AKT-mTOR, and JAK-STAT3, promoting cell growth, proliferation, and survival. PD-L1 upregulation following ROS1 fusion activation is relevant for immunotherapy.
