C-type lectin domain-containing protein 19A (CLEC19A) is a secreted protein belonging to the C-type lectin family, characterized by its carbohydrate-binding domain and potential ability to bind calcium ions[1][3]. The CLEC19A gene is predominantly expressed in brain tissue and is downregulated in glioma and glioblastoma, where overexpression has been shown to suppress tumor cell proliferation, reduce migration, arrest the cell cycle, and promote apoptosis in glioma cell lines and animal models[1]. Mechanistically, CLEC19A influences pathways including PI3K/AKT/NF-κB and regulates the expression of several genes related to tumor suppression and extracellular matrix remodeling. There is evidence for disease association with both aggressive periodontitis and a potential tumor suppressor role in brain tumors[1][3]. CLEC19A lacks a transmembrane domain, possesses an N-terminal signal peptide, and is believed to function extracellularly or within the secretory pathway[1][3]. Currently, it is emerging as a putative therapeutic target in oncology, specifically for glioblastoma, but no interacting drugs are documented[1][3].
