Overview

C-type lectin domain family 2 member L (CLEC2L) is a type II transmembrane cell surface protein belonging to the C-type lectin-like receptor (CTLR) superfamily, specifically the CLEC2 subfamily, which includes receptors such as CD69, LLT1, AICL, and KACL[1][2][4]. In contrast with most CLEC2 family members, which are broadly or hematopoietically expressed, CLEC2L displays predominantly brain-specific expression and is therefore also known as brain-associated C-type lectin (BACL)[1][4][7]. It is a non-glycosylated, disulfide-linked homodimer localized on the neuronal cell surface, especially in Purkinje cells of the central nervous system[4]. Unlike related CTLRs that function as immune receptors, current evidence does not support an immunological function for CLEC2L, and its precise physiological role in neurons remains uncharacterized[4]. No known drugs or clinical biomarkers are associated with CLEC2L, and it is not currently established as a therapeutic target in disease. **Note:** The gene is not encoded in the Natural Killer Gene Complex (NKC), unlike many related CTLRs. Its evolutionary conservation and unique tissue distribution suggest it may play as-yet-undiscovered roles in neurobiology[4].
