C-type lectin domain family 4 member C (CLEC4C), also known as BDCA2 or CD303, is a type II transmembrane glycoprotein selectively expressed on human plasmacytoid dendritic cells (pDCs). It functions as a receptor that binds non-sialylated galactose-terminated complex glycans. Upon ligand or antibody engagement, BDCA2 signals through the FcεRIγ chain, leading to a B-cell receptor-like signaling cascade and potent inhibition of Toll-like receptor-induced type I interferon (IFN-I) production by pDCs. This makes it an attractive therapeutic target for autoimmune diseases like systemic lupus erythematosus (SLE), where excessive IFN-I production is pathogenic.
