Overview

Immune cell pattern recognition receptors for beta-glucans, primarily represented by C-type lectin domain family 7 member A (Dectin-1), are critical sensors of the innate immune system that detect beta-1,3 and beta-1,6-linked glucans found in fungal cell walls and certain cereals. Upon binding these carbohydrate ligands, these receptors—which also include Complement Receptor 3 (CR3) and Toll-like receptors—initiate intracellular signaling pathways involving Syk kinase and Card9, leading to the activation of macrophages, dendritic cells, and neutrophils. This activation results in the production of pro-inflammatory cytokines, enhanced phagocytosis, and the induction of trained immunity, which provides a heightened state of readiness against subsequent infections. In the context of oncology, targeting these receptors with beta-glucan agonists is being explored to repolarize immunosuppressive tumor-associated macrophages into a tumoricidal M1-like phenotype. Additionally, these receptors play significant roles in the pathogenesis of inflammatory bowel disease and the host defense against invasive fungal infections like candidiasis and aspergillosis.

Mechanism of action

Agonism of pattern recognition receptors (primarily Dectin-1) on innate immune cells, triggering Syk-mediated signaling cascades that lead to cytokine production, phagocytosis, and enhanced antimicrobial/antitumor activity.
