Overview

C-type lectin domain family 7 member A, widely known as Dectin-1, is a type II transmembrane protein that serves as a critical pattern recognition receptor (PRR) on myeloid cells, including macrophages, dendritic cells, and neutrophils [11, 12]. It is primarily responsible for the recognition of beta-1,3-glucans, which are structural components of fungal cell walls and certain medicinal mushrooms like Grifola frondosa [10, 13]. Upon ligand binding, Dectin-1 signals through an immunoreceptor tyrosine-based activation motif (ITAM)-like domain to recruit Syk kinase, which subsequently activates downstream pathways such as NF-kB to induce the production of pro-inflammatory cytokines and enhance phagocytic activity [13, 15]. This process effectively bridges innate and adaptive immunity by promoting the maturation of antigen-presenting cells and the activation of cytotoxic T cells and natural killer (NK) cells [3, 12]. In the field of oncology, Dectin-1 is a therapeutic target for immunomodulatory agents like BLEX 404, a botanical beta-glucan extract [1, 2]. BLEX 404 acts as a Dectin-1 agonist to stimulate the immune system's anti-tumor response, particularly when used in combination with standard chemotherapy for conditions such as non-small cell lung cancer (NSCLC), triple-negative breast cancer, and pancreatic cancer [3, 4, 8]. Beyond its role in cancer immunotherapy, Dectin-1 is essential for host defense against fungal pathogens, and its dysregulation is linked to inflammatory bowel disease and other autoimmune disorders [10, 12]. Additionally, its ability to enhance hematopoiesis has led to clinical investigations for its use in treating myelodysplastic syndromes [4, 6].

Mechanism of action

Agonism; Dectin-1 activation triggers a Syk-dependent signaling cascade that activates NF-kB and MAPK pathways, leading to the production of pro-inflammatory cytokines (e.g., IL-12, TNF-alpha) and the stimulation of innate and adaptive immune cells [3, 11, 13].
