NKG2A is an inhibitory receptor expressed on NK cells and a subset of CD8+ T cells. It forms a heterodimer with CD94 and recognizes HLA-E, a non-classical MHC class I molecule. Engagement of NKG2A leads to recruitment of phosphatases that suppress activating signaling cascades, preventing inappropriate activation against healthy cells. NKG2A blockade or modulation has therapeutic potential for enhancing antitumor immunity or treating autoimmune diseases.
