CXCR1 and CXCR2 are closely related G protein-coupled receptors (GPCRs) that play key roles in immune cell signaling, particularly in the recruitment and activation of neutrophils. They bind CXC chemokines containing an ELR motif. Both receptors are expressed on neutrophils and are implicated in inflammation and cancer. CXCR1 is approximately 350 amino acids long and CXCR2 is slightly longer at about 355–360 amino acids. The two receptors share about 76% amino acid identity.
Ligand binding activates intracellular G proteins, leading to downstream signaling cascades that result in cellular responses such as migration, degranulation, respiratory burst activity, and increased cell survival. Blockade may reduce inappropriate inflammation or limit tumor progression by interfering with pro-tumorigenic signaling pathways.
