C-X-C motif chemokine ligand 10 (CXCL10) (CXCL10)

C-X-C motif chemokine ligand 10 (CXCL10), historically known as Interferon-gamma inducible Protein 10kDa (IP-10), is a potent pro-inflammatory chemokine belonging to the CXC family. It is primarily secreted by various cell types, including monocytes, endothelial cells, and fibroblasts, in direct response to Interferon-gamma (IFN-g) (UniProt P02778). CXCL10 functions as a specific ligand for the G protein-coupled receptor CXCR3, which is predominantly expressed on Th1-type T cells, natural killer (NK) cells, and macrophages. This interaction is critical for the directed migration and recruitment of these immune cells to sites of infection and chronic inflammation (NCBI Gene ID: 3627). Beyond its role in leukocyte trafficking, CXCL10 also possesses angiostatic properties, allowing it to inhibit neovascularization. In clinical contexts, elevated levels of CXCL10 are associated with the pathogenesis of autoimmune diseases such as rheumatoid arthritis and inflammatory bowel disease, making it a viable therapeutic target. Drugs like Eldelumab have been developed to neutralize CXCL10 to disrupt the cycle of chronic inflammatory cell recruitment (PMID: 25484347).
