CXCR2 is a G-protein-coupled receptor that binds multiple CXC chemokines, including IL-8, and plays a critical role in neutrophil recruitment, inflammation, and tissue homeostasis. It is expressed primarily on neutrophils, but also on oligodendrocyte lineage cells, basophils, T lymphocytes, and endothelial cells. Its activation triggers downstream signaling cascades such as PI3K/Akt, PLC/PKC, and MAPK/p38, leading to cell migration, proliferation, and survival. Dysregulation of CXCR2 is implicated in various diseases including neutrophilic inflammation, cancer progression, and neurological disorders, making it a therapeutic target for anti-inflammatory and neuroregenerative strategies.
CXCR2 antagonists block the binding of chemokines (e.g., IL-8) to the receptor, inhibiting downstream signaling pathways that mediate neutrophil recruitment and other inflammatory processes.
