Overview

C-X-C motif chemokine receptor 5 (CXCR5) is a transmembrane G protein-coupled receptor encoded by the CXCR5 gene on chromosome 11q23.3[1][3]. It primarily binds the chemokine ligand CXCL13 (BCA-1), mediating the targeted chemotaxis (migration) of B cells and subsets of T cells into lymphoid follicles where immune responses are organized[1][3]. CXCR5 is essential for B cell homing to lymph node follicles and the formation of germinal centers, as well as for enabling T follicular helper (Tfh) cells to support antibody production[3][8]. High expression of CXCR5 on B cells, Tfh cells, and certain lymphoma cells contributes to its utility as a biomarker and therapeutic target[1][3]. Overexpression has been linked to lymphoid cancers and certain solid tumors, where it may promote metastasis[3]. CXCR5 is under active investigation as a drug target in autoimmune diseases, cancer, and other immune-mediated conditions due to its central role in organizing adaptive immune responses and cell trafficking[8][2][3].
