Overview

C2 calcium-dependent domain-containing protein 4A (C2CD4A) is a nuclear protein characterized by the presence of a C2 domain, which is generally involved in calcium-dependent membrane interactions but can also function independently of calcium in some family members[1][3]. C2CD4A is implicated in regulating the acute inflammatory response, cell adhesion, and vascular permeability during inflammation, and is located in the nucleolus and nucleoplasm[2][3][5]. Genome-wide association studies have linked genetic variants in the C2CD4A region to type 2 diabetes and glycemic traits, particularly fasting blood glucose and insulin secretion; however, animal model studies suggest its loss alone may not disrupt glucose homeostasis due to functional redundancy[2]. C2CD4A is also implicated in cancer biology: its increased expression in colorectal cancer promotes tumorigenesis by inhibiting p53 signaling (via enhanced p53 ubiquitination), thereby facilitating cell growth and survival. Polymorphisms in C2CD4A and its paralog C2CD4B are associated with altered cancer susceptibility, including protective effects against lung cancer in some contexts[2]. No direct drug interactions, mechanisms of action, or validated clinical biomarkers have yet been associated with C2CD4A as a target.
