C2 domain-containing protein 2 (C2CD2) is a cytosolic and nuclear protein belonging to the C2 domain–containing family, with a still not well-defined molecular function[4][6]. C2CD2 and related proteins are involved in the regulation of lipid transport at membrane contact sites, notably at endoplasmic reticulum–plasma membrane junctions, contributing to calcium-dependent modulation of phosphoinositide signaling. This activity is critical for endocrine processes such as insulin secretion (characterized for its paralog TMEM24), and more broadly, for signal coordination in specialized cell types like pancreatic beta cells and neurons[1]. Genetic studies suggest its variants affect physical performance in aging (e.g., gait speed, grip strength) and it is implicated as a potential player in cancer biology through gene fusions and immune-related gene panels[1]. As of now, C2CD2 is not validated as a classical therapeutic target (receptor, enzyme, transporter, etc.), and there are no known drugs or mechanisms of action directly targeting this protein[1][3][4].
