C2 domain-containing protein 5 (C2CD5) is a calcium-dependent phospholipid-binding protein crucial for insulin-stimulated glucose transport, specifically required for the translocation of the glucose transporter SLC2A4/GLUT4 from intracellular vesicles to the plasma membrane in adipocytes. It is necessary for optimal vesicle fusion during this process. In neurons, C2CD5 is essential for proper mitochondrial trafficking and function, and its deficiency in animal models leads to hypothalamic mitochondrial abnormalities and obesity. C2CD5 is mainly classified as a vesicle trafficking protein with roles in calcium signaling and metabolic regulation. It is not currently considered a direct therapeutic target, and there are no drugs or approved biomarkers related to this molecule. Its aliases include CDP138, KIAA0528, and several variants referring to its molecular weight and phosphoprotein status.
