Overview

Cadherin-10 (CDH10) is a **type II classical cadherin**, a calcium-dependent cell adhesion molecule playing essential roles in mediating **homophilic cell-cell adhesion**. It is part of the cadherin superfamily, with a large N-terminal extracellular domain (composed of five subdomains, each with cadherin motifs), a single membrane-spanning region, and a conserved cytoplasmic tail[3][5]. CDH10 is **predominantly expressed in the brain**, where it contributes to synapse formation, axon guidance, and the maintenance of blood-brain barrier and specialized endothelial junctions[3][5]. It lacks the HAV (histidine-alanine-valine) recognition sequence seen in type I cadherins[3][5]. Mutations or loss of function in CDH10 have been implicated in various human cancers (notably lung squamous cell carcinoma, colorectal cancer, and pancreatic adenocarcinoma), with evidence for a role in tumor progression and metastasis[3][5]. Associations have also been noted with neurological diseases such as autism spectrum disorder[5]. No targeted therapies or approved drugs are currently known to interact selectively with CDH10, but its disruption marks altered cancer biology and possibly neurological function[3][5].
