Overview

Cadherin-24 (CDH24) is an unconventional type II cadherin involved chiefly in mediating strong calcium-dependent cell–cell adhesion[1][3]. The protein has five extracellular repeats containing calcium-binding motifs, a transmembrane domain, and a cytoplasmic tail that interacts with catenin proteins[1][2][3]. It is localized at cell–cell junctions and enables homophilic adhesion (binding identical molecules on adjacent cells), contributing to tissue architecture and organization. CDH24 binds to cytoskeletal regulatory proteins including alpha-, beta-, and delta-catenin, as characterized by Gene Ontology annotations[1][3]. Mutations or altered expression of CDH24 have been detected in certain disease contexts: for example, in gastric and colorectal cancers with high microsatellite instability, frameshift mutations in CDH24 may compromise its adhesion function, suggesting a tumor suppressor role[1]. Overexpression or altered expression has been identified in toxicological studies (e.g., in testicular injury under cadmium exposure), as well as in blood from twins discordant for psoriasis, implicating CDH24 in inflammatory diseases[1]. There are no documented drugs targeting CDH24 nor known clinical mechanisms of drug action. CDH24 does not currently have significant clinical use as a biomarker and there are no widely recognized safety or therapeutic concerns associated with this protein.
