Overview

Cadherin-4 (CDH4), also known as R-cadherin or retinal cadherin, is a classical cadherin from the cadherin superfamily and encodes a calcium-dependent cell-cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region, and a highly conserved cytoplasmic tail. CDH4 mediates homophilic and heterophilic interactions at cell-cell junctions, playing critical roles in neural development, brain segmentation, neuronal outgrowth, and the organization of retinal, kidney, and muscle tissues. It has been implicated in various pathological conditions, including cancer metastasis and certain congenital and developmental disorders. Although it is important in development and disease, no approved drugs are currently known to directly target Cadherin-4[1][2][3][9].
