Overview

Calcineurin-like phosphoesterase domain-containing protein 1 (CPPED1) is a human serine/threonine protein phosphatase and member of the calcineurin-like metallophosphoesterase superfamily. It specifically dephosphorylates AKT kinases at Ser-473, thereby acting as a negative regulator of the PI3K-AKT signaling pathway, blocking cell cycle progression, and promoting apoptosis. It may also play a role in inhibiting glucose uptake by adipocytes and be involved in chromatin remodeling as well as transcriptional regulation. CPPED1 is found in the cytosol and plasma membrane. Reduced protein levels have been noted in the placenta during term labor, suggesting additional regulation roles in pregnancy.

Mechanism of action

Inhibitors would be expected to potentiate PI3K-AKT signaling via maintained phosphorylation of AKT; activators or increased expression would be expected to suppress the pathway and promote apoptosis or cell cycle arrest. However, there are currently no known specific drugs targeting CPPED1
