Overview

The calcineurin pathway is a highly conserved calcium-dependent signaling cascade present in most eukaryotic organisms. The core of this pathway is the enzyme calcineurin—a serine/threonine protein phosphatase that is activated by calcium ions (Ca²⁺) and calmodulin. This pathway plays critical roles in immune response regulation, neuronal signaling, cardiac function, and fungal virulence. Targeting this pathway is crucial for preventing organ transplant rejection but also associated with side effects due to broad physiological roles.
