PPP3CA encodes the catalytic subunit alpha isoform of protein phosphatase 3, also known as calcineurin Aα. Calcineurin is a calcium/calmodulin-dependent serine/threonine protein phosphatase that plays a central role in transducing intracellular Ca²⁺-mediated signals. The enzyme is essential for various cellular processes, especially those involving calcium signaling. It controls phosphorylation states of many targets including synaptic receptors, transcription factors, ion channels, and cytoskeletal proteins. It plays critical roles in muscle differentiation—especially muscle fiber type conversion—and osteoclast/osteoblast regulation for bone formation.
