Calcineurin phosphatase is a calcium and calmodulin-dependent serine/threonine protein phosphatase that plays a central role in calcium-mediated signal transduction pathways. It is best known for its function in activating T cells of the immune system. Calcineurin is a heterodimer composed of a catalytic subunit (calcineurin A) and a regulatory subunit (calcineurin B). It is activated by an increase in intracellular Ca²⁺ concentration and binding of calmodulin. Calcineurin regulates numerous cellular processes through dephosphorylation events and is potently inhibited by immunosuppressive drugs such as cyclosporin A and tacrolimus (FK506).
