Calcipressin-3 (RCAN3) is a member of the RCAN (Regulator of Calcineurin) family of proteins, encoded by the *RCAN3* gene in humans. It is primarily known for inhibiting calcineurin, a calcium/calmodulin-dependent serine/threonine phosphatase, through direct binding to its catalytic domain. RCAN3’s expression is highest in brain and striated muscles, but it is also present in other tissues, including the heart and kidneys. By inhibiting calcineurin, RCAN3 modulates various calcium-dependent signaling pathways, impacting processes such as central nervous system development and cellular responses to stress. RCAN3 is also implicated in several diseases, particularly Down syndrome, where RCAN family proteins, including RCAN3, may contribute to the syndrome’s phenotype. Additionally, altered RCAN3 expression has been associated with various pathological conditions, including neurodegenerative diseases and metabolic disorders[1][2][3][5].
