Overview

The “calcitonin pseudogene” (CALCP, also called CALC3) refers to a member of the calcitonin gene family on human chromosome 11. Specifically, the CALC-III gene (referred to as CALC3) is a pseudogene: it contains only two exons that share homology with other calcitonin family genes (CALC-I, CALC-II), but it does not encode a functional calcitonin, calcitonin gene-related peptide (CGRP), or any other related peptide hormone. No protein translation has been observed from CALCP/CALC3; as such, it lacks biological activity, is not a “target” for drugs or therapeutic interventions, and is not implicated in any known physiological or pathological processes. The gene is considered functionally inert, with no known role as a biomarker or disease contributor[4]. The calcitonin gene family contains several genes (CALC-I to CALC-V), of which only some are functional. CALC-III (CALC3, CALCP) is identified as a pseudogene and not a source of calcitonin or CGRP in humans[4]. There is no evidence that CALCP encodes a receptor, enzyme, transporter, or any molecular target, nor does it play a role in disease or drug mechanisms. CALCP/CALC3 is a noncoding pseudogene, not a valid drug target, and its mention as a receptor or target is incorrect based on genomic and functional data[4].
