The calcium-activated non-selective cation channel (CAN channel), most notably represented by the TRPM4 protein, is a membrane ion channel that opens in response to increased intracellular calcium (Ca2+) levels. It is permeable to monovalent cations such as sodium (Na+), potassium (K+), and cesium (Cs+), but not to Ca2+ itself. TRPM4 is a tetrameric protein with multiple transmembrane and cytosolic domains. Intracellular ATP inhibits channel activity. TRPM4 is expressed in various tissues, including cardiac tissue and immune cells, where it plays roles in cardiac pacemaking, immune cell migration, and exocrine secretion.
