Calcium and integrin-binding family member 4 (CIB4) is a small, EF-hand domain-containing protein that binds calcium and magnesium ions[3][4]. This binding increases its structural stability. It belongs to the CIB family, which also includes CIB1, CIB2, and CIB3, all of which are evolutionary conserved and share similar structures[2][3]. CIB4’s specific biological functions are not as well-studied as CIB1 or CIB2, which play established roles in integrin signaling, the cardiovascular system, and hearing[2]. Its main biochemical properties are ion binding and potential modulation of protein-protein interactions, especially with integrin family members. In other species, such as sheep, CIB4 interacts with proteins involved in calcium-mediated signal transduction, including calcineurin subunits and plasma membrane calcium pumps[1]. However, its expression is more limited, and there are no established disease associations or drug interactions for CIB4 in humans[2][4].
