Overview

Calcium-binding protein 1 (CABP1) is a calcium-binding, neuron-specific regulatory protein belonging to the calmodulin superfamily. It contains four EF-hand motifs and modulates activity of multiple calcium channels and receptors, including L-type and P/Q-type voltage-gated calcium channels, inositol 1,4,5-triphosphate receptors, and TRPC5 channels[1][2][4][5]. CABP1 regulates neuronal calcium signaling, thereby influencing synaptic transmission, cellular excitability, and protection against excitotoxic degeneration. It exists in several isoforms (e.g., caldendrin), with differential subcellular localization in neurons and sensory cells[2]. CABP1 expression is highest in the retina and brain, notably in the hippocampus, cortex, amacrine, and cone bipolar cells[1][2]. While not a direct therapeutic target, CABP1 has been implicated in brain tumor prognosis and may influence neurological disease due to its role in calcium homeostasis[2][4].
