Overview

Calcium-binding protein 39 (CAB39), also known as MO25, is a conserved scaffold protein that functions as a core component of the LKB1–STRAD–MO25 complex, which is required for the stable activation and proper localization of the serine/threonine kinase LKB1[1][2][3][4]. Through this complex, CAB39 plays a critical role in stabilizing LKB1 and facilitating downstream signaling, notably the activation of AMP-activated protein kinase (AMPK) and pathways involved in cellular metabolism, energy sensing, and stress response. CAB39 also participates in regulating serine/threonine phosphorylation and is a target of regulatory microRNAs that can modulate cellular metabolic signaling. Aberrant CAB39 expression or regulation has been linked to pathologies including metabolic disease, certain cancers, and cardiovascular dysfunctions. There are no well-established drugs that directly target CAB39 as of current evidence, and it is not widely considered a canonical therapeutic target[1][2][3][4][5].
