Overview

Calcium-binding protein 39-like (CAB39L) is a member of the calcium-binding protein family, functioning primarily as a scaffolding/adaptor protein that modulates intracellular signal transduction. CAB39L forms part of a protein complex with STK11 (also known as LKB1) and STRAD, supporting the catalytic activity and stability of serine/threonine kinases such as LKB1, which is key for AMPK pathway activation[7]. CAB39L plays important roles in cell metabolism—especially in preventing the Warburg effect in gastric cancer by activating the LKB1-AMPK-PGC1α pathway, acting as a tumor suppressor[1]. In addition, CAB39L contributes to cytoskeletal rearrangement, cell motility, and possibly to renal salt transport via its effects on kinase signaling complexes. CAB39L expression is subject to regulation via promoter methylation and may serve as a prognostic biomarker in certain cancers[1]. Diseases associated with CAB39L include monocytic leukemia and ovarian squamous cell carcinoma[7]. No known drugs directly targeting CAB39L have been described, but its potential as a therapeutic target—particularly in metabolic and neoplastic diseases—has been identified.
