Overview

Calcium-binding protein 5 (CaBP5) is a neuronal, calmodulin-like protein characterized by multiple EF-hand calcium-binding motifs. It is predominantly expressed in the retina, particularly in the inner nuclear layer, suggesting roles in visual signal transduction and synaptic function[4][3][2]. CaBP5 modulates calcium-dependent cellular processes, including the inhibition of L-type calcium channel inactivation and regulation of vesicle exocytosis and neurotransmitter release[4][3]. It interacts with proteins such as Munc18-1 and myosin VI, both involved in synaptic vesicle cycling, and may influence neurite outgrowth and organization[3][4]. While mutations or disruptions in related CaBP family members have been linked to various neuronal and sensory disorders, CaBP5 itself has not been established as a direct therapeutic target or biomarker, though its dysfunction may contribute to visual system diseases such as fundus dystrophy[4]. There is some risk of confusion with CaBP3, a distinct but related protein, as some aliases overlap[4][2]. In summary, CaBP5 is a modulator of calcium signaling, critical for retinal and neuronal function, but is not currently a recognized therapeutic target or biomarker in clinical practice[4][3][2].
