Overview

Calcium-binding proteins and effectors represent a broad functional class of molecules that translate intracellular calcium signals into specific biological responses [IUPHAR, 2024]. This group includes calcium sensors, such as calmodulin and the S100 protein family, which undergo conformational changes upon binding calcium via specialized motifs like the EF-hand [UniProt, 2024]. These sensors then interact with and regulate various effector proteins, including kinases (e.g., CaMKII) and phosphatases (e.g., calcineurin), to control vital processes such as muscle contraction, neurotransmitter release, and gene transcription [StatPearls, 2023]. Dysregulation of these pathways is a hallmark of several pathologies, including heart failure, neurodegenerative diseases like Alzheimer's, and various cancers [PubMed, 2022]. Therapeutically, these proteins are significant; for instance, calcineurin is the target of major immunosuppressants like cyclosporine, while troponins serve as gold-standard biomarkers for cardiac injury [NCBI, 2023]. However, the ubiquitous nature of calcium signaling poses a challenge for drug development, necessitating high specificity to avoid systemic toxicity and off-target effects [PubMed, 2023].
