Overview

Calcium channels are a diverse class of membrane proteins that facilitate the selective movement of calcium ions into cells, serving as a bridge between electrical signals and biological responses. They are broadly divided into voltage-gated calcium channels (VGCCs), which are triggered by membrane depolarization, and ligand-gated calcium channels (LGCCs), which are activated by chemical messengers like glutamate (StatPearls, 2023). These channels play pivotal roles in physiological processes such as cardiac and smooth muscle contraction, the release of neurotransmitters at synapses, and the regulation of gene transcription (UniProt, 2024). In clinical medicine, VGCC blockers like amlodipine and diltiazem are cornerstone treatments for hypertension and supraventricular arrhythmias (PubMed, 2022). Meanwhile, LGCCs like the NMDA receptor are therapeutic targets for managing Alzheimer's disease and treatment-resistant depression (NIH, 2023). Because calcium is a ubiquitous second messenger, the dysregulation of these channels is implicated in numerous diseases, including chronic pain, epilepsy, and various cardiovascular disorders.

Inhibition of calcium ion influx through the channel pore or modulation of the channel's gating properties to reduce intracellular calcium concentrations and dampen cellular excitability.
