The calcium channel protein alpha 1 subunit is the principal pore-forming component of voltage-gated calcium channels (VGCCs), responsible for the selective influx of Ca2+ ions into cells upon membrane depolarization. This process is critical for numerous physiological functions, including muscle contraction, neurotransmitter release, gene expression, and cardiac action potentials. There are ten known genes encoding alpha 1 subunits in humans, grouped into L-type, P/Q-type, N-type, R-type, and T-type channels, each with distinct tissue distribution and function. It determines most biophysical properties of VGCCs including ion selectivity, gating kinetics, voltage sensitivity, and pharmacological profile. Accessory subunits modulate trafficking and gating. Alpha 1 subunits are targeted by clinically important drugs for cardiovascular and neurological disorders. Mutations in specific genes are linked to various diseases.
