This group encompasses voltage-gated calcium channels (VGCCs), which open in response to membrane depolarization, and various exchangers like the sodium-calcium exchanger (NCX) and the plasma membrane Ca2+ ATPase (PMCA) that maintain calcium homeostasis (UniProt). These proteins are essential for critical physiological functions, including cardiac and smooth muscle contraction, neurotransmitter release, and the activation of calcium-dependent signaling pathways (StatPearls). In clinical practice, they are primary targets for treating cardiovascular conditions such as hypertension and arrhythmias, as well as neurological disorders like epilepsy and chronic pain (PubMed, PMID: 30285334). Drugs targeting these proteins, such as calcium channel blockers (CCBs), work by inhibiting ion flow to reduce vascular resistance or dampen overactive neuronal firing (StatPearls).
