Target intelligence / Profile preview
Calcium channels and exchangers are a broad class of transmembrane proteins that facilitate and regulate the flux of calcium ions (Ca2+) across biological membranes (IUPHAR/BPS Guide to Pharmacology). This group encompasses voltage-gated calcium channels (VGCCs), which open in response to membrane depolarization, and various exchangers like the sodium-calcium exchanger (NCX) and the plasma membrane Ca2+ ATPase (PMCA) that maintain calcium homeostasis (UniProt). These proteins are essential for critical physiological functions, including cardiac and smooth muscle contraction, neurotransmitter release, and the activation of calcium-dependent signaling pathways (StatPearls). In clinical practice, they are primary targets for treating cardiovascular conditions such as hypertension and arrhythmias, as well as neurological disorders like epilepsy and chronic pain (PubMed, PMID: 30285334). Drugs targeting these proteins, such as calcium channel blockers (CCBs), work by inhibiting ion flow to reduce vascular resistance or dampen overactive neuronal firing (StatPearls).
Inhibition of voltage-gated calcium channels to reduce calcium entry into cells, leading to vasodilation or reduced neuronal excitability; modulation of sodium-calcium exchangers to influence cardiac contractility (StatPearls, IUPHAR/BPS).
