Calcium deficiency, clinically known as hypocalcemia, is a pathological state characterized by insufficient levels of calcium in the extracellular fluid or body stores (StatPearls, 2023). Calcium is a critical divalent cation that serves as a secondary messenger in signal transduction and is essential for the structural integrity of the skeleton, neuromuscular function, and enzymatic activities (NIH, 2024). The condition often arises from vitamin D deficiency, hypoparathyroidism, or renal failure, leading to symptoms ranging from muscle cramps and paresthesia to life-threatening arrhythmias and seizures (Mayo Clinic, 2023). While calcium deficiency is a clinical diagnosis rather than a single molecular target, its management involves pharmacological intervention at the Calcium-sensing receptor (CaSR) and Vitamin D receptor (VDR) to restore homeostasis (PubChem, 2024). Therapeutic strategies primarily focus on oral or intravenous calcium supplementation and the administration of vitamin D analogs to enhance intestinal absorption (StatPearls, 2023).
Pharmacological management involves direct ion replacement to restore serum levels and the use of Vitamin D receptor agonists to increase intestinal calcium absorption and renal reabsorption.
